Clinical trial design affects interpretation of trial results.

Clinical trial designs have evolved over time to reflect our increasing knowledge of tumor biology. For example, basket trials examine the effect of a drug on multiple tumor types that share a common genetic mutation. Alternatively, umbrella trials enroll patients with a single tumor type who are matched to treatment strategies tailored to the presence of particular genetic aberrations. Furthermore, adaptive trials address some of the inefficiencies of traditional clinical trials by allowing modification of the trial design based on interim data. Understanding the complexities of clinical trial design and interpretation is foundational toward providing patients with optimal care.
